چکیده

The traditional anti-convulsant drug Valproic Acid (VPA) has been found to be involved in suppressing cancer progression while modulating various cancer-associated signaling pathways. In particular, VPA acts as either a Histone Deacetylase (HDAC) inhibitor or a Notch signaling activator in suppressing tumor growth. VPA is less toxic, and by itself, has limited anti-tumor effects. Thus, VPA has been used as an adjuvant in combination with a variety of other anti-cancer agents for many types of cancers. These combination strategies display potential applications in cancer treatments. In particular, VPA could up-regulate certain G Protein-Coupled Receptors (GPCRs) in some cancer cells. Some of these GPCRs are highly expressed naturally in many cancer cells and these characteristics have been applied towards novel enhanced combination therapeutics with VPA and specific receptor-targeted cytotoxic peptide-drug conjugates. *Corresponding author: Lichun Sun, Professor, Department of Medicine, Peptide Research Laboratories, Tulane Health Sciences Center, New Orleans, LA 701122699, USA, Tel: 504-988-1179; Fax: 504-988-3586; E-mail: [email protected] Received January 02, 2014; Accepted January 22, 2014; Published January 27, 2014 Citation: Sun L, Coy DH (2014) Anti-Convulsant Drug Valproic Acid in Cancers and in Combination Anti-Cancer Therapeutics. The multifaceted therapeutic potential of valproic acid beyond CNS axis

Valproic acid is widely used as an anti-epileptic globally. Anti-epileptic action is mediated by Gamma Amino Butyric Acid (GABA) receptors. In past and recently, several other clinical outcomes have been proposed by various studies. These under-reported clinical actions apart from anti-epileptic activity are mediated by various intracellular and extracellular pathways. Several mechanisms like H...

The correlation between telomerase activity and Bax/Bcl-2 ratio in valproic acid-treated MCF-7 breast cancer cell line

Objective(s):Valproic acid (VPA), a drug used in the treatment of neurological disorders, has been shown to have cytotoxic effects on cancer cells through different mechanisms. Telomerase, a ribonucleoprotein reverse transcriptase, is responsible for elongation of the telomere and is activated in cancers. A relation between telomerase activity and resistance to apoptosis has been established. T...

Evaluation of Anti-cancer activity of Boswellic acid and Montelukast sodium against human prostate cancer cell line PC-3

Prostate cancer is a devastating disease for which current therapies are inadequate. Various lines of evidences have suggested the 5-lipoxygenase (5-LOX) pathway and the leukotriene receptor pathway are potential targets for prevention or treatment of Prostate cancer. Thus, search for new anti-cancer drugs targeting 5-LOX and leukotriene is very essential and important. The objective of the pre...
